The connection between ALS and inflammation is strengthened by the finding that TDP-43 interacts with the p65 subunit of the inflammatory protein NF-kB p65 in the spinal cords of ALS patients. Researchers propose that TDP-43 and p65 act together to influence microglia cells to initiate an inflammatory response that may lead to the death of spinal cord neurons. In a mouse model of ALS, treatment with an agent capable of blocking p65 activity minimized neuron loss and eased disease symptoms.
